Hypertension is a disorder which afflicts one-third of all American adults and one-half of the elderly. Hypertension is strongly associated with coronary artery disease and cerebrovascular disease and with increased risk of heart attack and stroke. In the United States there are over 15 million affected, the rate of disease increases with age and hypertension disproportionately affects African Americans. This proposal seeks to better understand the complex interactions between the kidney and the vasculature, especially the intracellular mechanisms of sodium homeostasis in the vascular smooth muscle cell. The first aim of this proposal is to investigate the effects of paracrine regulation of these ion transport mechanisms by epithelial cells and to develop a model for the regulation of apical Na/H exchange in the vascular smooth muscle cell. In the second aim, we will examine the regulation of transport by changes in intracellular Ca. We will focus on the mechanism of Ca influx through the L-type channel which has been shown to be altered in some hypertension models. Finally, we will examine the effects of the sympathetic nervous system and hormones on ion transport mechanisms in vascular smooth muscle cells. The third aim will examine the control of G protein-mediated transport in renal cells. The fourth aim will test the hypothesis that Ca/calmodulin kinase II (CaM kinase II) is the protein that phosphorylates a cytoplasmic linker protein linking the Na/K-ATPase to the membrane. This research is relevant to the prevention of hypertension since new treatments could be directed to the specific targets within the vascular cell and renal epithelial cell, thus increasing the safety of these agents in order to diminish their side effects.package ddospro
