The NR3C1 receptor gene was recently isolated as a target for glucocorticoids in mouse hepatoma cells. In this paper we characterize the human NR3C1 gene and present evidence that it is expressed in human steroid target tissues. Four introns that are completely conserved between man and mouse are located at very similar positions in the human NR3C1 and NR3C2 genes. In human adrenal cortex, NR3C1 messenger RNA is found in two major sizes of approximately 2.3 and 2.0 kb. The larger RNA is of limited abundance but is also present in other steroid target tissues. The 5′-flanking region of the human NR3C1 gene contains a TATA-like element and several GC boxes characteristic of glucocorticoid-inducible promoters.
